Abstract:
ObjectiveTo onstruct recombinant adenovirus vector pDC316-hIL-24 and detect the viral titer.MethodsTotal mRNA was ex tracted from HEK293 cells, and specific primer pairs were designed in order to clone human interleukin-24 (hIL-24) gene cDNA by RT-PCR method. hIL-24 gene was directional constructed into shuttle plasmidr (pDC316-EGFP) after digestion by restrictive endoenzyme BglⅡ, and HindⅢ. HEK293 cells were co-transfected with the constructed recombinant shuttle plasmid pDC316-EGFP and large adenovirus helper plasmid pBHGlox (delta) E1, 3Cre in mediation of liposome. The constructed recombinant adenovirus vector was named pDC316-hIL-24. Recombinant adenovirus was amplificated and purificated in HEK293 ce lls.ResultsRecombinant adenovirus vector pDC316-hIL-24 was constructed successfully and high titer recombinant adenovirus was obtained.ConclusionThe construction of recombinant adenovirus vector pDC316-hIL-24 and the obtain of recombinant adenovirus will lay down the basis for developing gene therapy of cancer.

Abstract:
ObjectiveTo investigate the characterization of anti-Aβ autoantibodies in AD patients and affinity ability of autoantibodies stainning the Aβplaques in vitro.MethodsELISA was performed to detect levels of naturally occuring anti-Aβautoantibodies in surum from AD patients, vascular dementia and age-macthed healthy controls. Immunohistochemistry assay was employed to confirm the affinity abillity of anti-Aβautoantibodies stainning the Aβ plaques in vitro.ResultsNaturally occuring anti-Aβautoantibodies in surum from AD patients was significantly lower than those from vascular dementia and healthy controls. The autiantibodies levels were independent of gender, age and MMSE scores in AD patients. Autoantibodies purified from healthy individuals and vascular dementia patients could readily stain the Aβplaques in transgenic mouse Tg2576, while those from AD patients could not.ConclusionCompared with vascular dementia and healthy controls, AD patients have obviously lower anti-Aβautoantibodies which could not stain Aβplaques in transgenic mouse and showed a low-affinity ability with Aβplaques in vitro. The petential pathophysiological function of autoantibodies in AD patients needs futher study.

Abstract:
ObjectiveTo study the transvaginal three-dimensional ultrasound (3--DTVS) in the diagnosis of endometrial carcinoma and to judge muscular infiltration depth value.Methodsfrom January 2010 to June 2010 in our hospital after surgical treatment of 148 patients with endometrial cancer. All cases of preoperative application of more than a 3-DTVS plane and vascular imaging and measurement function diagnosis of endometrial carcinoma and to evaluate muscular infiltration depth (3-DTVS group); 45 cases by nuclear magnetic resonance (MRI) examination as control group (MRI), the coincidence rate with pathological results after surgical judgment, compared two groups of diagnosis of endometrial carcinoma and muscular infiltration.ResultsThrough 3-DTVS diagnosis coincidence rate of carcinoma of endometrium was 90.1%, the judgment of muscular infiltration coincidence rate was 78.3%. Application 3 DTVS with magnetic resonance imaging (MRI) diagnosis of endometrial carcinoma and muscular infiltration accuracy there was no significant difference between the two groups (P >0.05).Conclusion3-DTVS in diagnosis of endometrial carcinoma and muscular infiltration depth has important diagnostic value.

Abstract:
ObjectiveTo compare dynamic change of nasal resistance before and after ATD SCIT.Method43 cases of adult patients with allergic rhinitis, tracking ATD SCIT, recording symptom and sign scores、nasal resistance before and15 weeks after treatment, 1 year after treatment. Compare symptom and sign scores, nasal resistance all periods.Resultsymptom and sign scores (single symptom scores, total symptom scores)、nasal resistance before dust mites SCIT and 15 weeks after treatment, 1 year after treatment was statistically significant (F=318.811, P < 0.001). Nasal resistance with symptom and sign scores is no correlation (r=0.77, P>0.5).Conclusionnasal resistance gradually improve with allergic rhinitis immunotherapy, but no correlation with symptom and sign scores, continuous dynamic detection of nasal resistance can assess effect objectively, can be used to supple the symptom and sign scores.

Abstract:
ObjectiveTo investigate the clinical value of interventional embolization in huge hemangioma of auricle and to analyse its curative effects.MethodRetrospective analysis about 5 cases of auricle large hemangioma in recent 3 years, preoperative line all patients underwent digital subtraction angiography (DSA), and were treated by intervention embolization successfully. The tumor shape, intraoperative bleeding, wound healing and complications were observed.ResultAfter preoperative vascular embolization, 5 cases tumors had become smaller and darker. Within 48 h, 5 cases underwent surgical treatment. Less intraoperative bleeding (about 100 ml per case on average), well wound healing and no complications such as postoperative bleeding and hemiplegia.ConclusionThe preoperative using of interventional embolization showed an excellent clinic effect on treatment of huge hemangioma of auricle.To huge hemangioma of auricle with rich blood supply, preoperative embolization could be reduce the operative bleeding efficiently, shorten the operation and improve the successful rate of operation.

Abstract:
ObjectiveTo evaluate the effect of transvenous embolization treatment of refractory traumatic carotid-cavernous fistula (TCCF), and emphasize the importance of keeping carotid artery unobstuction.MethodsWe retrospectively analyzed 8 TCCF treated by transvenous coil combined Onyx glue embolization approach in the past three years. all cases treated by transarterial balloon blocking the fistula approach failed, then received transvenous embolization of the fistula. After 3 -18 months, we evaluated the efficiacy through cerebral angiography and clinical follow-up.Results5 cases treated by simple transvenous embolization completely blocked the fistula. 2 cases treated by transarterial detachable balloon combined with transvenous embolization completely blocked the fistula, the cure rate was 87.5%. 1 case treated by transvenous embolization appears the cavernous pseudoaneurysm, the aneurysm disappeared after three months. None of the patients reported distal cerebral embolism and cerebral bleeding during operation, but 2 cases first appear abducens nerve palsy, gradually improved and eventually disappeared after 2-6 months. After follow-up 3-18 months, fistula occlusion of 7cases completely blocked and carotid artery unobstructed. Fistula of 1case disappeared, the ipsilateral carotid artery obstructed. Eye symptoms and intracranial murmur of all cases disappeared.ConclusionTransvenous embolization treatment of traumatic carotid-cavernous fistula is safe and effective. To preserve the internal carotid artery, it can be used as an important method for treating refractory TCCF

Abstract:
ObjectiveTo investigate the changes of immunity and life quality after the treatment of cytokine induced-killer cells (CIKs) for patients with advanced NSCLC after chemotherapy, and to assess the effectiveness and safety of CIKs treatment.MethodSixty-one patients with advanced NSCLC after chemotherapy treated with CIKs were enrolled as CIKs group, and another sixty-one post-chemotherapy patients of advanced NSCLC treated with optimal supportive care during the same period were enrolled as control group. Before and after CIK cells being transfused back, the changes of immune system including subsets of T cells, quality of life (Karnofsky score), therapeutic effects and side effects were compared between the two groups.ResultCompared with before CIK cells therapy, the ratios of CD3+and CD3+/CD4+T cells significantly increased (P < 0.05), while there was no significant change in the control group. KPS score significantly increased in the CIKs group, which was 67.27±8.94(P < 0.05). The disease control rate (DCR) were 85.25% in the CIKs group, which was significantly higher than those in the control group (59.01%)(P < 0.05). The major adverse events were transient fever and skin rash, and no other side effect was observed.ConclusionCIK cells treatment is safe and effective for advanced NSCLC patients. It can improve the immune imbalance, DCR and quality of life of the patients.

Abstract:
ObjectiveTo summarize the experience in minimally invasive treatment of pectus carinatum.MethodsThe clinical data of 32 pectus carinatum patients who underwent minimally invasive reverse Nuss technique in our hospital from July 2010 to July 2013 was retrospectively analyzed, with 26 males and 6 females aging (11.8±6.7) years. The Haller index (HI) was 2.06±0.13. The pectus carinatum was corrected by placing steel bar subcutaneously anterior to the sternum to apply pressure to the sternum, and the steel bar was fixed to the ribs through plates.ResultsAll the 32 cases received operation successfully, with no death occurring in the perioperative period. Operation time was (67.0±7.8) min, blood loss was (25.0±5.2) ml, and hospital stay was (6.0±2.8) d. The thoracic appearance returned to normal after operation and the HI was 2.7±0.4. The thoracic appearance and chest CT image were satisfying. Follow-up investigation of the 32 cases was performed for 1-36 months, 18 months on average. The steer bar was removed in 4 cases. Excellent effect was observed in 27 cases (84.4%) and good effect was observed in 5 cases (15.6%).ConclusionModified minimally invasive reverse Nuss technique is a simple, safe and effective surgical method to correct the pectus carinatum of older patients, which has good recent effect.

ObjectiveTo discuss the Mediterranean anemia patients and the Mediterranean anemia patients blood test report in RBC histogram and indexes of blood platelet histogram correlation.MethodsStatistics in January 2005 to December 2012 our hospital outpatient hospital patients fully automatic blood cell analyzer blood test report all test results of 200 cases with Mediterranean anemia gene, Combined with Mediterranean anemia gene test results, Divided into two groups, A group for the diagnosis of the Mediterranean anemia blood test report 100 cases of patients, Another group of Mediterranean anemia gene detects the Mediterranean anemia in 100 cases of normal patients blood test report, Blood test results of two groups of patients of red blood cell and platelet histogram graph compares histogram graphics.ResultsThe Mediterranean anemia patients blood test results of platelet histogram presents obvious bimodal characteristics of 82 cases; Platelet histogram of bimodal characteristics of 18 cases; Red blood cells obviously single-peak histograms left 79 cases, No shift to the left of the 21 cases. Gene detects the normal of the Mediterranean anemia patients blood test report platelet histogram shows the bimodal characteristics of 8 cases of obvious; Platelet histogram of bimodal characteristics of 92 cases; 10 cases of RBC histogram unimodal apparent shift to the left, 90 cases without a shift to the left. The comparison results compare similar between the two groups was statistically significant (P < 0.05).ConclusionsFrom the blood test report of observing platelet histogram and histogram of red blood cells morphological characteristics. Through the blood routine test results can provide a rapid screening of Mediterranean anemia and reliable clinical diagnosis basis.

ObjectiveTodiscuss causes and characteristics of acid-related upper gastrointestinal bleeding of elderly people in emergency department. Methods 239 patients with acid related gastrointestinal bleeding in the elderly were analyzed retrospectively.ResultsElderly acid-related upper gastrointestinal bleeding accounted for 57.87% of upper gastrointestinal bleeding, which accounted for 1.67% of gastroesophageal reflux disease, accounting for 63.18% of peptic ulcer, gastritis or duodenal inflammation accounted for 9.21%, accounting for 0.84% of functional dyspepsia, accounted for 25.10% of acute gastric mucosal lesion. 27 died, one died of bleeding in 1 case directly, accounting for 3.70%, chronic systemic disease progression and died of bleeding complications and multiple organ failure secondary to 24 cases, 88.89%. 11.30% of the total mortality.ConclusionThe acid-related diseases are the main reason for upper gastrointestinal bleeding in the elderly, prognostic factors and systemic underlying diseases tend to worsen the situation.

ObjectiveTo explore the expression and clinical significances of CD4+CD29+T cells for patients with acute spinal cord injury (ASCI).Methodcollecting 75 cases of patients with spinal cord injury, with 50 cases of healthy people as control group, using flow cytometry test peripheral blood CD4+CD29+ helper T cells and intracellular cytokines gamma interferon (IFN-gamma) and interleukin 4(IL-4) content; In 3 and 7 days after injury, review three and analyze the correlation between the three and Frankel classification.Resultscompared with normal, spinal cord injury patients with CD4+T cells and IL-4+CD29 content increased significantly, but the content of IFN-gamma significantly decreased (P < 0.05);Treatment for 3 days, CD4+T cells and IL-4+CD29 is injury to some extent on day 1, and IFN-gamma recovery in different level, of which the change of D and E significantly (P < 0.05).Treatment for 7 days, CD4+T cells and IL-4+CD29 continue to fall, IFN-gamma continue to rebound, D and E+T cells in patients with CD4+CD29, IFN-gamma and IL-4 than day3 have significant difference (P < 0.05).Correlation analysis suggests CD4+T cells and IL-4+CD29 with Frankel classification has significant positive correlation, and IFN-gamma and Frankel classification has significant negative correlation.ConclusionCD4+CD29+helper T cells and IL-4, and IFN-gamma reduce is one of the important immune characteristics of spinal cord injury associated with disease outcomes, and and relevance to the damage degree, therefore has important clinical significance.

Micrornas (miRNAs) are a class of endogenous non-coding small RNA, length of about 18~25 nucleotides, which combined with specific mRNA target for inhibition of mRNA translation process, so as to adjust to change the expression level of target gene at the post-transcriptional level and closely related to the occurrence, development, invasion and metastasis of malignant tumor.Previous researches have shown that DNA methylation may be the regulatory mechanisms of abnormal expression of micrornas in breast cancer, Including micrornas low methylation activation of oncogene and high methylation deactivation of tumor suppressor genes which can lead to abnormal expression of downstream target genes and participate in the development of breast cancer.Therefore, this article were mainly reviewed on regulation of DNA methylation of micrornas in breast cancer.

ObjectiveTo assess the effectiveness and safety of vitamin K1 in prevention of bleeding for cirrhotic patients.MethodsComputerized searches were conducted to retrieve randomized controlled trials (RCTS) comparing vitamin K1 to placebo in prevention of bleeding for cirrhotic patients from PubMed、EMbase、Cochrane Library、CNKI、VIP、WANGFANG、CBM databases.Data collection by Inclusion criteria.RevMan5.0 was used for statistical analysis.Adverse drug reactions of vitamin K1 were summarized.ResultsWe could not find any randomized trials on vitamin K1 in prevention of bleeding for cirrhotic patients; 569 cases of adverse reactions of vitamin K1 were analyzed, cirrhosis in which accounts for 11.9%.ConclusionThe necessary of vitamin K1 to prevent bleeding in cirrhotic patients remains large scale and multicenter RCTS to further confirmed.The experts reach a consensus that using vitamin K1 for 3 days on patients of obstructive jaundice, intestinal dysfunction, and those taking long-term warfarin therapy or using antibiotics that hinder the absorption and metabolism of vitamin K, can restore the the reserves of vitamin K.If patients with liver disease fail in the use of vitamin K1, the treatment cannot be continued.At this moment, infusion of fresh frozen plasma, cryoprecipitate, prothrombin complex or fibrinogen can be considered according to the coagulation tests.

ObjectiveTo research the effect on the integration of management based on community and family with psychopaths of community rehabilitation.MethodsBring the support factors of family into three level of management pattern including mental heath hospital of city-the hospital of town-village clinics at this time, increase the important effect on the support factors of family, collect and manage data changes of the psychopaths before and after getting social (family) support, the quality of the comprehensive life, social harmfulness and other cases.ResultAfter implementing the integration of management based on community and family, the psychopaths'obtained the acceptance and care from family (the ratio of care increased from 42.24% to 77.37%).The quality of the comprehensive life had significant improvement (At the beginning, the average of the total points of the quality of life questionnaire was 111.21, and then descended to 107.96, the lower of the score, the better of the quality of life).It also reduced the relapse of making-trouble (The relapse of making-trouble was reduced from 9.91% to 4.09%.It shows that the difference was statistically significant (P < 0.05).ConclusionThe integration of management based on community and family (pay attend to the participation of the family factor), it can significantly improve the psychopath's quality of life and reduce the relapse of making-trouble.
