ObjectiveTo explore the relationship between expression level of aquaporin-4 (AQP4), protein kinase C (PKC) and brain edema based on the focal cerebral ischemia reperfusion rat model. MethodsThe rat model of middle cerebral artery (MCA) was duplicated by suture method .The rats were randomly divided into normal control group, sham-operated group and cerebral ischemia reperfusion(CIR) group. Water permeability for rat astrocytes was evaluated by the green fluorescent protein(GFP) labeled AQP4 tracing system. Scores of neurofunctional defect were graded and water content in brain tissues was measured. Expression of AQP4 and PKC in brain tissue was detected by immunohistochemistry. ResultsNeurofunctional defect occurred at 6 h after CIR. Water content in brain tissue increased at 12 h after CIR, reached peak at day 1-3. The expression levels of AQP4 were decreased first, then increased gradually at 12 h later, obviously increased at 24 h after CIR and reached to highest point at day 3. While PKC increased gradually at 6 h after CIR, kept high level for 5 days. ConclusionBrain edema may relate to the rise of PKC and AQP4, PKC may reduce the expression of AQP4 via phosphorylation in the early stage of CIR in order to delay the process of brain edema.

Abstract:
Ubiquitin proteasome system is physically ess ential to normal cell functions. Fbw7 is a subunit of E3 ligase specifically binding to a series of substrates to promote their lysosomal degradation, the regulation of which is commonly mutated in tumors. Most substrates of Fbw7 are dominant oncogenes, including Cyclin E, c-Myc, Notch, Aurora A, and Mcl1, which intensively regulate cell cycle, DNA repair, metastasis as well as chemoresistance. Therefore, Fbw7 targeting intervention can inhibit malignant transformation of cells. This review focuses on the biological functions, genetic alterations and regulation networks of Fbw7 in order to highlight its role in tumor progression.

Abstract:
Myocardial infarction is the most common cause that induces heart failure. Recent studies have identified that instant and prolonged activation of inflammatory response is a key character of post-infarction heart failure. Inflammatory response to some degree can promote wound healing, scar formation and recovery of ischemic myocardium, but excessive inflammation could play an important role in the development of chamber dilation, systolic dysfunction and heart failure. Extensive necrosis in the infarcted myocardium triggers the innate immune response through stimulating Toll-Like Receptor (TLR) mediated signaling, activating the complement cascade and reactive oxygen species (ROS) pathway, and regulating the Nuclear Factor (NF)- κB system. Neutrophils, mononuclear cells, macrophages and other kinds of cells play essential roles in inf lammation. How to control inflammation to suppress pathological remodeling is an important issue to be considered in developing new treatment for heart failure. This review summarizes the roles of related cells and signal pathways in the pathophysiological mechanisms of inflammation in myocardial induced heart failure in order to provide references for further study in the effects of inflammation and anti-inflammatory therapies in heart failure.

Abstract:
In recent years, the discovery of Kisspeptins makes people have a new understanding of the neuroendocrine axis regulating puberty and reproduction. Kisspeptins is not only the upstream factor that promotes the secretion of GnRH, but also regulates the hypothalamic pituitary ovary axis, which is mainly manifested in the regulation of gonadotropin secretion, the initiation of puberty and the regulation of reproductive function. By observing the healthy population and reproductive endocrine disorders, we found that Kisspeptins can stimulate the secretion of GnRH, which induces LH secretion, and enhance the pulse frequency of LH. At present, many animal experiments have proved that Kisspeptins has the potential function in the following aspects, such as the function of ovary, embryo implantation and placenta formation. This article will focus on the existing basis for the Kisspeptins regulation of GnRH pulse type secretion, in particular, the initiation of puberty and reproductive endocrine aspects.
