Research progress of CD11c+ cells in inflammatory skin diseases

摘要: CD11c是主要分布于髓样细胞表面的一种跨膜糖蛋白,与炎症反应、免疫应答以及免疫细胞的活化、转化和增殖等一些重要的病理过程密切相关。CD11c+细胞在各种炎症性皮肤病的真皮层血管周围、淋巴结内大量聚集提示CD11c+细胞不单纯是由局部细胞产生的,很可能是是由其他细胞产生并通过全身的淋巴-血液循环运输到炎症部位并参与皮肤局部炎症反应的。CD11c+细胞的这种参与作用又与T细胞(Th1/Th2/Th17)、角质形成细胞之间有着密不可分的联系。一方面,CD11c+细胞可通过影响T细胞的分化来参与皮肤炎症反应。皮损处不同表型CD11c+DCs的激活是决定特应性皮炎向Th2/Th1免疫应答发生的关键。银屑病皮损处CD11c+DCs可以诱导活化的Th1/Th17细胞分泌促炎细胞因子INF-γ/IL-17。角质形成细胞通过表达人胸腺间质淋巴细胞生产素, 可以激活CD11c+DCs, 进而诱导Th2型免疫应答的发生。另一方面,CD11c+DCs通过分泌IL-36诱导角质形成细胞分泌各种细胞因子和趋化因子, 进一步作用于角质形成细胞使其过度增生从而加剧皮炎反应。Abstract: CD11c is a transmembrane glycoprotein mainly distributed on the surface of myeloid cells. Tt is closely related to inflammatory response, immune response and immune cell activation, transformation and proliferation and some other important pathological process. CD11c+cells accumulate in large quantities around the dermal vessels and the lymph nodes.It suggests that CD11c+cells are not simply produced by local cells and are likely to be produced by other cells and transported through the systemic lymphatic-blood circulation to participate in the local inflammatory response. The involvement of CD11c+cells in a variety of inflammatory skin diseases is inextricably related to T cells (Th1/Th2/Th17) and keratinocytes.On one hand, CD11c+cells can participate in the skin inflammatory by affecting the differentiation of T cells.The activation of different phenotype CD11c+DCs is the key to determine the AD to Th2 / Th1 immune response.CD11c+ DCs in psoriatic lesions can induce activated Th1 / Th17 cells to secrete proinflammatory cytokine INF-γ / IL-17.Keratinocytes express human thymus mesenchymal lymphocyte production hormone, which can activate CD11c+ DCs and induce the Th2 immune response.On the other hand, secretion of IL-36 by CD11c +DCs induces keratinocytes to secrete various cytokines and chemokines, further acting on keratinocytes to hyperproliferation and thereby exacerbating dermatitis.
