The research of the foldable capsular vitreous body (FCVB) in mechanically releasing nerve growth factor

目的通过折叠式人工玻璃体(FCVB)对小鼠神经生长因子(NGF)在体外缓释作用的研究,为FCVB能否建成眼内药物缓释系统提供初步实验依据。 方法体外实验在透皮仪中进行,将4 mL浓度为3 μg/ml的NGF注入FCVB,浸没于含15 mL无菌水的小烧杯中,收集30 min、1、2、4、8和12 h FCVB外的缓释液,用分光光度法制作NGF溶液标准曲线,并检测样品中NGF的浓度,吸收波长定为280 nm。 结果实验数据显示,FCVB能够缓释NGF,并且缓释浓度随时间的延长而增长。 结论FCVB能够在体外物理缓释NGF,并且缓释浓度呈时间依赖性增长。此研究为进一步研究FCVB能否建成眼内药物缓释系统提供依据,同时为临床相应疾病的治疗提供帮助。 Abstract:Objective To investigate the role of the foldable capsular vitreous body (FCVB) in mechanically releasing nerve growth factor (NGF). Our study supplies the experimental evidences for further investigation contributing in intraocular drug deliver systerm exploration. Methods We choose in vitro study. NGF sustained-release studies in vitro was finished in cups of modi fied Franz diffusion cells. Concentration of 3 ug/ml NGF diluted in steril water was injected into the FCVB capsules, immersed into small cup with 15 ml steril water. Two hundred (200) μl liquid was aspirated at time intervals of 30 min, 2, 4, 8, 12 h separatively. Standard curve was made through ultraviolet spectrophotometry examination. The assumption length of NGF examination was 280 nm. Results The data from in vitro study indicated that FCVB could mechanically release NGF in dose and time dependent manner. At 12h time point, NGF was released at concentration of 465ng/ml. Conclusions Data from in vitro study showed that FCVB could sustainably, mechanically release NGF via capsule apertures in a time-dependent and dose-dependent manner. This research for the further study can confirm FCVB built intraocular drug sustained-release system to provide the basis for clinical at the same time corresponding treatment of diseases. -
